Virulence Phenotyping of Leaf Rust (Puccinia triticina) Isolates from Southern Pakistan

Collections of Puccinia triticina were made from farmers’ fields five different agro-ecological locations (Sakrand, Tandojam, Larkana, Sanghar and Badin) Sindh province, Pakistan 2015 2016, to identify the virulence variation. Single uredinial isolates investigated for phenotyping on 24 near isogenic (Thatcher wheat) lines which differ single Lr resistance genes. Spores two (Sakrand Tandojam) not viable could be revived only urediniospores three (Larkana, revived. None pathotypes had Thatcher wheat with leaf rust genes Lr23 Lr42. However, Lr24, LrB, Lr10, Lr14b Lr20 exhibited susceptibility response i.e. (HITs 3 4) all tested pathotypes. Based virulence, ten phenotypes (MSCTNS, RTSTNS, RKTRGS, PNDQDS, JDBQGJ, MDPSDS, RTPTPS, MNPSDS, MJLTGS MSPTDS) identified among isolates, designated six-letter code. Two RTSTNS RTPTPS broad spectrum, both virulent nineteen while pathotype JDBQGJ narrow spectrum as compared other tested, just eight rust. Among variability was also recorded. Most races more than one Resistance (Lr42 Lr23) effective can exploited achieve in wheat. Further, study provides profile area may help manage pathogen.
